目的探讨姜黄素对前列腺癌PC-3细胞热休克蛋白27(HSP27)表达的影响。方法体外培养人前列腺癌PC-3细胞,随机将其分为4组,空白对照组不给药;大剂量组给予50μmol/L姜黄素2μl;小剂量组给予25μmol/L姜黄素2μl;阴性对照组给予等体积二甲基亚砜(DMSO)。采用免疫荧光染色检测PC-3细胞HSP27的表达情况,采用RT-PCR检测PC-3细胞HSP27 m RNA的表达情况。结果免疫荧光染色结果显示,姜黄素治疗组PC-3细胞HSP27染色强度明显低于空白对照组和阴性对照组,并且大剂量组HSP27染色强度降低更加明显,与空白对照组比较,差异有统计学意义(P<0.05)。RT-PCR分析结果显示,姜黄素治疗组PC-3细胞HSP27 m RNA的表达明显降低(50%和60%),且呈剂量依赖性,与空白对照组比较(128%),差异有统计学意义(P<0.05)。结论姜黄素可抑制PC-3细胞HSP27的表达,可能是其抗前列腺癌的机制之一。
Objective To investigate the effect of curcumin on the expression of heat shock protein 27 (HSP27) in PC-3 prostate cancer cells. Methods Human prostate cancer PC-3 cells were cultured in vitro and divided into 4 groups randomly. The blank control group was given no drug. The high dose group was given 2μmol / L curcumin. The low dose group was given 25μmol / L curcumin. The negative control Groups were given equal volume of dimethyl sulfoxide (DMSO). The expression of HSP27 in PC-3 cells was detected by immunofluorescence staining. The expression of HSP27 mRNA in PC-3 cells was detected by RT-PCR. Results Immunofluorescence staining showed that the staining intensity of HSP27 in PC-3 cells treated with curcumin was significantly lower than that in blank control group and negative control group, and the intensity of HSP27 staining in high-dose group was more obvious than that in blank control group Significance (P <0.05). The results of RT-PCR showed that the expression of HSP27 mRNA in PC-3 cells was significantly decreased (50% and 60%) in the curcumin-treated group compared with the blank control group (128%), the difference was statistically significant Significance (P <0.05). Conclusion Curcumin can inhibit the expression of HSP27 in PC-3 cells and may be one of its mechanisms of anti-prostate cancer.
