目的探讨凋亡抑制基因Survivin和增殖细胞核抗原(PCNA)在自然杀伤(NK)/T细胞淋巴瘤中的表达及其关系。方法NK/T细胞淋巴瘤标本25例,经过组织学形态及CD45RO、CD3ε、CD20免疫组织化学分析,免疫组织化学链霉素抗生物素-过氧化酶连接法(SP法)检测T细胞内抗原(TIA-1)、粒酶B,原位杂交法检测EB病毒编码的mRNA(EBER 1/2),按照2000年WHO新的淋巴瘤分类方法明确诊断。应用SP法检测25例NK/T细胞淋巴瘤组织和10例反应性增生淋巴组织中的Survivin和PCNA表达水平,分别计数1 000个肿瘤细胞中Survivin及PCNA阳性细胞所占百分比,作为Survivin阳性率及细胞增殖指数(PI)。结果25例NK/T细胞淋巴瘤组织中Survivin表达阳性17例(68.0%),10例反应性增生淋巴组织中Survivin不表达,其差异有统计学意义(P<0.01);25例NK/T细胞淋巴瘤PI为(41.48±5.10)%,10例反应性增生淋巴组织PI为(20.10±2.77)%,二者比较差异有统计学意义(t=12.39,P<0.01);Survivin表达阳性组PI为(43.35±4.80)%,显著高于阴性组[(37.50±2.90)%](t=3.16,P<0.01);Survivin表达与平均PI呈正相关(r=0.637,P<0.01)。结论NK/T细胞淋巴瘤中Survivin基因和PCNA表达上调,Survivin可能通过调控细胞增殖参与NK/T细胞淋巴瘤的发生发展。
Objective To investigate the expression of survivin and proliferating cell nuclear antigen (PCNA) in natural killer (NK) / T-cell lymphoma and their relationship. Methods Twenty - five NK / T cell lymphomas were studied by histological examination and immunohistochemical staining of CD45RO, CD3ε, CD20 and immunohistochemical streptavidin - peroxidase (SP) (TIA-1), granzyme B and Epstein-Barr virus (EBER 1/2) were detected by in situ hybridization. The diagnosis was made according to the 2000 WHO classification of new lymphoma. The expression of Survivin and PCNA in 25 cases of NK / T cell lymphoid tissue and 10 cases of reactive hyperplasia lymphoid tissue was detected by SP method. The percentage of Survivin and PCNA positive cells in 1 000 tumor cells were counted as the positive rate of Survivin And cell proliferation index (PI). Results The positive expression of Survivin in 25 cases of NK / T cell lymphomas was 17 cases (68.0%). Survivin was not expressed in 10 cases of reactive hyperplasia lymphoid tissues, the difference was statistically significant (P <0.01); 25 cases of NK / T The PI of cell lymphoma was (41.48 ± 5.10)% and that of 10 cases of reactive hyperplasia lymphoid tissue was (20.10 ± 2.77)%, the difference was statistically significant (t = 12.39, P <0.01) PI was (43.35 ± 4.80)%, which was significantly higher than that in the negative group (37.50 ± 2.90)% (t = 3.16, P <0.01). Survivin expression was positively correlated with the mean PI (r = 0.637, P <0.01). Conclusions Survivin gene and PCNA expression are up-regulated in NK / T cell lymphoma. Survivin may be involved in the development of NK / T cell lymphoma by regulating cell proliferation.
