作者介绍一种新的计数人全血T淋巴细胞的细菌玫瑰花技术(即大肠杆菌玫瑰花试验),采血量少,简易,快速且不需特殊设备。将真空冻干的大肠杆菌(ATCC 11303)(S-igma)接种于硫羟乙酸盐培养基中培养24小时。用0.15MNaCl洗后,在10%甲醛(PH7.2的磷酸盐缓冲盐水配制)中固定过夜,用盐水充分洗涤,然后置分光光度计取650nm波长,以盐水将细菌浓度调整至光密度15。此悬液可保存数月。试验时,取肝素抗凝血100ul,用Hanks 液洗一次,低速离心,吸去
The authors introduce a new bacterial rose blood cell counting technique for human whole blood T lymphocytes (E. coli Rose test), which has low blood collection, is simple, fast and requires no special equipment. Lyophilized E. coli (ATCC 11303) (S-igma) was inoculated into thiolacetate medium and cultured for 24 hours. After washing with 0.15M NaCl and fixing in 10% formaldehyde (pH7.2 phosphate buffered saline) overnight, washing well with brine and then setting the spectrophotometer to 650nm, the concentration of bacteria was adjusted to optical density 15 in saline.
