目的 筛选人源抗HIV 1gp12 0趋化蛋白受体结合位点噬菌体Fab单克隆抗体基因。方法 根据HIV 1gp12 0趋化蛋白受体结合位点的氨基酸序列合成 2 3肽 ,并以此为固相抗原从HIV 1噬菌体Fab抗体库筛选阳性克隆 ,并进行鉴定及序列测定。结果 获得了 1株抗HIV 1gp12 0趋化蛋白受体结合位点的人源Fab抗体克隆 ,具有较高的亲和力、特异性和抑制率 ,序列测定及分析显示该抗体属IgGⅠ亚类 ,κ型 ,重、轻链可变区分别属VhⅢ和VκⅢ基因家族。结论 成功地获得了人源抗HIV 1gp12 0趋化蛋白受体结合位点噬菌体Fab单克隆抗体基因。
Objective To screen the Fab monoclonal antibody gene of human anti-HIV 1gp12 0 chemokine receptor binding site phage. Methods According to the amino acid sequence of HIV 1 gp12 0 chemotactic protein receptor binding site, 23 peptides were synthesized and used as solid phase antigen to screen positive clones from HIV 1 phage Fab antibody library. The positive clones were identified and sequenced. Results A clone of human Fab antibody against HIV 1gp12 0 chemokine receptor binding site was obtained with high affinity, specificity and inhibition rate. Sequence analysis and analysis showed that the antibody was of IgG subclass, The heavy and light chain variable regions belong to VhⅢand VκⅢgene families, respectively. Conclusion Human monoclonal anti-HIV 1gp12 0 chemokine receptor binding site phage Fab monoclonal antibody gene was successfully obtained.
