目的:明确激素性股骨头坏死(SANFH)大鼠模型形成过程中骨组织Ca、P、Mg、Zn元素含量的变化规律。方法:96只SD大鼠随机分为空白组与模型组,每组48只,采用尾静脉注射大肠杆菌脂多糖40μg/kg,2d后臀肌注射甲基强的松龙40mg/kg,3d,建立SANFH模型;空白组臀肌注射等体积0.9%氯化钠溶液。分别于造模4、8、12周后分批处死各组大鼠,分离股骨,通过系统方法学考察建立股骨中各元素电感耦合等离子体发射光谱法(ICP-OES)含量测定方法,分析比较SANFH大鼠模型形成过程中股骨各元素含量变化。结果:各元素在其线性范围内均呈现良好的线性关系(r≥0.999),平均加样回收率在99.96%-101.40%之间(RSD<3%,n=6)。与空白组比较,模型组股骨Ca、P元素含量均呈下降趋势,但雄性模型组大鼠股骨Ca、P含量在造模8、12周显著下降(P<0.05);雌性模型组大鼠股骨P含量在造模8周显著性下降(P<0.05);而模型组大鼠股骨Mg、Zn含量则无显著差异。结论:SANFH发病过程中会伴随引起骨组织Ca、P元素含量的变化,提示可将股骨Ca、P含量作为SANFH模型评价指标及药物干预的药效评价指标。
Objective: To determine the regularity of Ca, P, Mg and Zn contents in bone during the formation of steroid-induced osteonecrosis of the femoral head (SANFH) rat model. Methods: Ninety-six Sprague-Dawley rats were randomly divided into blank group and model group with 48 mice in each group. The mice were injected with 40μg / kg Escherichia coli lipopolysaccharide via caudal vein, 40mg / kg methylprednisolone, Establish SANFH model; blank group gluteal injection volume 0.9% sodium chloride solution. Rats in each group were sacrificed at 4, 8, and 12 weeks after modeling. Femurs were isolated and the content of ICP-OES in the femur was determined by systematic method. Changes of femur elements content during the formation of SANFH rat model. Results: The linearity of each element showed good linearity (r≥0.999), and the average recovery was between 99.96% and 101.40% (RSD <3%, n = 6). Compared with the blank group, the contents of Ca and P in the femur of the model group showed a decreasing trend. However, the content of Ca and P in the femur of the model group decreased significantly at 8 and 12 weeks (P <0.05) The content of P in the model group decreased significantly (P <0.05) at 8 weeks, while there was no significant difference in the content of Mg and Zn in the model group. CONCLUSIONS: The changes of Ca and P contents in bone tissue accompanied with SANFH pathogenesis suggest that the content of Ca and P in femur may be used as the evaluation index of SANFH model and the efficacy evaluation index of drug intervention.
