目的建立测定大鼠血浆中COH-05[(Z)-5-(3-氨基-4-甲氧基苯基)-4-(3,4,5-三甲氧基苯基)-3H-1,2-二硫基-3-酮]质量浓度的HPLC法,并应用该法研究COH-05在大鼠体内的药物动力学。方法色谱柱为Kromasil C18柱(250 mm×4.6 mm,5μm),流动相为乙腈-水-甲酸(体积比为60.0∶40.0∶0.2),检测波长为245 nm,同系物COH-07作为内标,测定大鼠血浆中COH-05的质量浓度。结果 COH-05的线性范围为0.02~2.50 mg.L-1(r=0.995 8),定量下限为0.02 mg.L-1,回收率为88.6%~92.8%,日内和日间精密度均小于8.5%。结论 HPLC法适用于COH-05血药浓度测定及药物动力学研究。
Objective To establish a method for the determination of COH-05 [(Z) -5- (3-amino-4-methoxyphenyl) -4- (3,4,5-trimethoxyphenyl) -3H-1 , 2-dithianyl-3-one] HPLC method was used to study the pharmacokinetics of COH-05 in rats. Methods The chromatographic column was Kromasil C18 column (250 mm × 4.6 mm, 5 μm). The mobile phase was acetonitrile-water-formic acid (volume ratio 60.0:40.0:0.2). The detection wavelength was 245 nm and the homologue COH-07 was used as internal standard , The concentration of COH-05 in rat plasma was measured. Results The linear range of COH-05 was 0.02-2.50 mg.L-1 (r = 0.995 8). The lower limit of quantification was 0.02 mg.L-1. The recoveries were 88.6% -92.8%. The intra- and inter-day precision were less than 8.5%. Conclusion HPLC method for the determination of COH-05 blood concentration and pharmacokinetics.
