目的建立高效液相色谱法测定小鼠血浆及组织样品中氟尿嘧啶(5-Fu)的浓度,并进行5-Fu在小鼠体内的药物动力学研究。方法以5-溴尿嘧啶(5-Bru)为内标,血浆和组织样品经液-液萃取后,采用SHIMADZU高效液相色谱仪进行分析,色谱柱为Kromasil C18(4.6 mm×200 mm,5μm),流动相为乙腈-水(1︰99,V︰V),检测波长为265 nm。结果该法测定5-Fu在血浆及各组织中的平均回收率较高,日间、日内精密度符合有关规定。结论方法操作简便、快速、适用于5-Fu在小鼠血浆、组织中的检测及药物动力学研究。
Objective To establish a method for the determination of 5-fluorouracil (5-Fu) in mouse plasma and tissue samples by high performance liquid chromatography (HPLC) and to study the pharmacokinetics of 5-Fu in mice. Methods 5-Bromine (5-Bru) was used as an internal standard. The plasma and tissue samples were analyzed by liquid-liquid extraction using SHIMADZU HPLC. The chromatographic column was Kromasil C18 (4.6 mm × 200 mm, 5 μm ). The mobile phase consisted of acetonitrile-water (1: 99, V: V) with a detection wavelength of 265 nm. Results The method of determination of 5-Fu in plasma and each tissue average recovery rate is high, day, day precision in line with the relevant provisions. Conclusion The method is simple, rapid and suitable for the detection and pharmacokinetics of 5-Fu in mouse plasma and tissues.
