目的:探讨抗人小扁豆凝集素结合型甲胎蛋白异质体单克隆抗体(AFP-R-LCAMcAb)对原发性肝癌细胞的特异性亲和力及放射免疫显像。方法:应用(131)Ⅰ标记的AFP-R-LCAMcAb对11例AFP阳性肝癌、6例AFP阴性肝癌及4例肝炎后肝硬化患者进行放免显像,观察不同时间肝癌显像情况。结果:11例AFP阳性肝癌患者中有9例肿瘤区显像,而6例AFP阴性肝癌及4例肝炎后肝硬化患者无1例出现肝肿瘤区及肝内局部显像。且小剂量(131)I-AFP-R-LCAMcAb对人体肝功能及凝血酶原时间无明显影响(P<0.05)。结论:AFP-R-LCAMcAb对人AFP阳性肝癌细胞有较强的特异性亲和力,有希望成为肝癌放免检测及治疗的理想载体。
Objective: To investigate the specific affinity and radioimmunoscintigraphy of anti-human lentils lectin-binding alpha-fetoprotein heteroantibody (AFP-R-LCAMcAb) on primary hepatocellular carcinoma cells. METHODS: The 131I-labeled AFP-R-LCAMcAb was used to perform radioimmunoassay on 11 cases of AFP-positive HCC, 6 cases of AFP-negative HCC, and 4 cases of post-hepatitis cirrhosis. The imaging of HCC at different times was observed. RESULTS: Nine out of 11 patients with AFP-positive liver cancer had tumor area imaging, while none of 6 patients with AFP-negative liver cancer and 4 patients with post-hepatitis cirrhosis had liver tumor area and intrahepatic local imaging. Low-dose (131)I-AFP-R-LCAMcAb had no significant effect on human liver function and prothrombin time (P<0.05). Conclusion: AFP-R-LCAMcAb has strong specific affinity for human AFP-positive hepatocellular carcinoma cells. It is hopeful that AFP-R-LCAMcAb will be an ideal carrier for detection and treatment of liver cancer radioimmunoassay.
