目的:研究复方六月青(CLYQ)对肝星状细胞(HSC)I型胶原(Col I)合成的影响。方法:采用四甲基噻唑蓝(MTT)法检测CLYQ对细胞的毒性,分别采用免疫细胞化学法和实时荧光定量PCR法检测CLYQ对HSC的Col I蛋白和mRNA表达的影响。结果:CLYQ对HSC毒性很低。CLYQ各剂量组中HSC的Col I蛋白表达显著减弱(P<0.01)。CLYQ高、中剂量组均可显著下调HSC的Col I mRNA表达(P<0.01)。结论:CLYQ通过下调HSC的Col I蛋白及mRNA表达,从而减少细胞外基质(ECM)合成,发挥抗肝纤维化作用。
Objective: To study the effect of CLYQ on the synthesis of type I collagen (Col I) in hepatic stellate cells (HSC). Methods: The cytotoxicity of CLYQ was detected by MTT method. The effects of CLYQ on the expression of Col I protein and mRNA of HSC were detected by immunocytochemistry and real-time fluorescence quantitative PCR respectively. Results: CLYQ has low toxicity to HSC. Col I protein expression of HSC in each dose group of CLYQ was significantly weakened (P <0.01). Both CLYQ high and middle dose groups could significantly down-regulate Col I mRNA expression in HSC (P <0.01). Conclusion: CLYQ can reduce the extracellular matrix (ECM) synthesis and decrease the hepatic fibrosis by down-regulating the expression of Col I protein and mRNA in HSC.
