目的:探讨ω-3脂肪酸对SW480结肠癌干细胞增殖及血管内皮生长因子(VEGF),syndecan-1,Ki-67基因表达的影响。方法:用流式细胞仪分选出SW480细胞中的CD133+/CD44+/ESA+细胞亚群(SW480结肠癌干细胞),将结肠癌干细胞分用系列浓度(15,30,60μg/μL)的EPA(二十碳五烯酸),系列浓度(15,30,60μg/μL)的DHA(二十二碳六烯酸)及溶剂处理(对照组),用MTT法检测各组细胞的增殖情况;并用RT-PCR检测DHA(60μg/μL)或EPA(60μg/μL)处理后结肠癌干细胞VEGF,syndecan-1和Ki-67表达的变化。结果:低浓度(15μg/μL)的DHA和EPA对结肠癌干细胞的生长无明显影响(均P>0.05),中、高(30,60μg/μL)浓度的EPA和DHA能明显抑制结肠癌干细胞的生长,与对照组比较差异均有统计学意义(均P<0.05)。60μg/μL的EPA及DHA能明显抑制syndecan-1和Ki-67基因的表达,与对照组比较差异均有统计学意义(均P<0.05),但两者对VEGF基因的表达无明显影响(均P>0.05)。结论:ω-3脂肪酸可抑制SW480结肠癌干细胞的增殖,其机制可能与抑制syndecan-1和Ki-67的表达有关。
Objective: To investigate the effects of omega-3 fatty acids on the proliferation of SW480 colon cancer stem cells and the expression of VEGF, syndecan-1 and Ki-67. Methods: The CD133 + / CD44 + / ESA + cell subsets (SW480 colon cancer stem cells) in SW480 cells were sorted by flow cytometry. The colon cancer stem cells were divided into five groups (DHA (docosahexaenoic acid)) and solvent treatment (control group) at different concentrations (15, 30 and 60 μg / μL). The proliferation of each group was detected by MTT assay. The expression of VEGF, syndecan-1 and Ki-67 in colon cancer stem cells treated with DHA (60μg / μL) or EPA (60μg / μL) RESULTS: DHA and EPA at low concentration (15μg / μL) had no significant effect on the growth of colon cancer stem cells (all P> 0.05). EPA and DHA at medium and high concentrations (30 and 60μg / μL) significantly inhibited colon cancer stem cells Compared with the control group, the differences were statistically significant (all P <0.05). 60μg / μL EPA and DHA significantly inhibited the expression of syndecan-1 and Ki-67, which were significantly different from the control group (all P <0.05), but both had no significant effect on the expression of VEGF gene All P> 0.05). CONCLUSION: Omega-3 fatty acids can inhibit the proliferation of SW480 colon cancer stem cells, which may be related to the inhibition of the expression of syndecan-1 and Ki-67.
