目的研究人骨髓源性成骨细胞的生物学特性,为骨组织工程选择理想的种子细胞提供依据。方法采用全骨髓培养法培养骨髓基质干细胞,传代扩增后采用诱导培养基进行诱导,对骨髓基质干细胞诱导成的成骨细胞进行细胞形态学观察,检测碱性磷酸酶活性及分泌钙结节和I型胶原的能力。结果骨髓基质干细胞可以进行传代和大量扩增,经诱导培养后能够诱导出成骨细胞,诱导出的成骨细胞具有典型成骨细胞的形态学和生物学特性。结论骨髓源性成骨细胞具有良好的成骨细胞特性,可以作为骨组织工程的种子细胞。
OBJECTIVE: To study the biological characteristics of human bone marrow-derived osteoblasts and provide the basis for selecting ideal seed cells for bone tissue engineering. Methods Bone marrow stromal stem cells were cultured by whole bone marrow culture method. After passaged and expanded, the cells were induced by induction medium. The morphology of osteoblasts induced by bone marrow stromal stem cells was observed. The activity of alkaline phosphatase and the secretion of calcium and Type I collagen capacity. Results Bone marrow stromal stem cells could be passaged and expanded in large quantities. After induction, osteoblasts could be induced. The induced osteoblasts had the morphological and biological characteristics of typical osteoblasts. Conclusion Bone marrow-derived osteoblasts have good characteristics of osteoblasts and can be used as seed cells for bone tissue engineering.
