目的明确非雌激素依赖型(Ⅱ型)和雌激素依赖型(Ⅰ型)子宫内膜癌在基因表达上的差异,找寻非雌激素依赖型子宫内膜癌的恶性标志物。方法利用全基因组Affymetrix基因芯片技术对2003年10月至2005年10月南方医科大学南方医院妇产科收治的3例雌激素受体(ER)和孕激素受体(PR)阳性及3例ER和PR阴性子宫内膜癌患者的子宫内膜癌标本进行研究,检测FOSB、NTRK2、JUN等基因在非雌激素依赖型中的表达情况,结合生物信息学分析,并采用半定量逆转录聚合酶链反应(RT-PCR)验证结果。结果在非雌激素依赖型子宫内膜癌中FOSB、NTRK2和JUN基因明显高表达。结论FOSB、NTRK2和JUN基因或许可以作为非雌激素依赖型子宫内膜癌的恶性标志物。
Objective To investigate the difference in gene expression of non-estrogen-dependent (type II) and estrogen-dependent (type I) endometrial carcinoma and to find out the malignant markers of non-estrogen-dependent endometrial carcinoma. Methods The genome-wide Affymetrix microarray technique was used to detect the expression of three ER and PR positive and three ER in the Department of Obstetrics and Gynecology, Nanfang Hospital, Southern Medical University from October 2003 to October 2005. And PR-negative endometrial cancer patients with endometrial cancer specimens to detect FOSB, NTRK2, JUN and other genes in non-estrogen-dependent expression, combined with bioinformatics analysis and the use of semi-quantitative reverse transcription polymerase Chain reaction (RT-PCR) validation results. Results The expression of FOSB, NTRK2 and JUN genes was significantly higher in non-estrogen-dependent endometrial carcinoma. Conclusion FOSB, NTRK2 and JUN genes may be used as malignant markers of non-estrogen-dependent endometrial cancer.
