目的探讨结肠癌组织中白细胞蛋白酶抑制因子(SLPI)的表达及其临床意义。方法选择150例结肠癌及癌旁正常组织制成微阵列芯片作为研究对象,用免疫组织化学方法检测组织芯片中SLPI蛋白表达并评分,采用χ2检验比较不同SLPI表达水平和结肠癌临床病理性特征,包括年龄、性别、组织分化程度、TNM分期、淋巴结转移及远处转移之间的关系。结果 SLPI在结肠癌组织中相对于正常结肠组织高表达,SLPI蛋白高表达与肿瘤分化[低分化:8(42.1%)vs 11(57.9%);中/高分化:3(2.3%)vs128(97.7%)、TNM分期:[Ⅲ~Ⅳ:25(29.4%)vs 60(70.6%)]、Ⅰ~Ⅱ:2(3.1%)vs 63(96.9%)、淋巴结转移[18(28.6%)vs 45(71.4%)]及远处转移[11(84.6%)vs 2(15.4%)]相关(P<0.05),而与患者的性别和年龄差异无统计学意义(P>0.05)。结论 SLPI表达水平与肿瘤分化程度、TNM分期、淋巴结转移及远处转移有关,有可能作为判断结肠癌患者预后的指标。
Objective To investigate the expression of leukocyte protease inhibitor (SLPI) in colon cancer and its clinical significance. Methods 150 cases of colorectal cancer and adjacent normal tissue were selected as the microarray. The expression of SLPI protein in tissue microarray was detected by immunohistochemistry and the expression of SLPI protein was compared with Chi-square test, and the clinicopathological features of colorectal cancer , Including the relationship between age, gender, histological grade, TNM stage, lymph node metastasis and distant metastasis. Results SLPI was overexpressed in colorectal cancer tissues compared with that in normal colon tissues. The expression of SLPI protein was significantly correlated with tumor differentiation (poorly differentiated: 8 (42.1%) vs 11 (57.9%); moderate / well differentiated: 3 97.7%), TNM stage: [Ⅲ ~ Ⅳ: 25 (29.4%) vs 60 (70.6%)], Ⅱ ~ 2: 3.1% vs 63.96% 45 (71.4%)] and distant metastasis [11 (84.6%) vs 2 (15.4%)] (P <0.05), but not with the gender and age of the patients (P> 0.05). Conclusion The expression level of SLPI is related to the degree of tumor differentiation, TNM stage, lymph node metastasis and distant metastasis, which may be used as an index to judge the prognosis of patients with colon cancer.
