目的 :观察普罗布考 (probucol)对血管平滑肌细胞 (VSMCs)核因子κB(NF-κB)活性的调控作用 ,以探讨 probucol在动脉粥样硬化 (AS)和经皮腔内冠状动脉成形术后再狭窄 (RS)防治中的某些可能的作用机制。 方法 :采用电泳迁移位移测定法观察 probucol对 VSMCs NF-κB活性的影响。结果 :H2 O2 或胎牛血清 (NCS)处理 VSMCs 72 h后 ,NF-κB活性均明显增强 ;而加入 10 0 μm ol/ L probucol后 ,NF- κB活性受到部分抑制 ,抑制率分别为 37.1%和 14.8%。 结论 :Probucol抑制 NCS、H2 O2 诱导的 VSMCs NF- κB的活性 ,可能是其临床有效防治 RS及 AS的作用机制之一
OBJECTIVE: To observe the regulatory effect of probucol on the activity of nuclear factor kappa B (NF-κB) in vascular smooth muscle cells (VSMCs) in order to investigate the role of probucol in the pathogenesis of atherosclerosis (AS) and percutaneous transluminal coronary angioplasty Some possible mechanisms of action in the prevention and treatment of restenosis (RS). Methods: The effect of probucol on NF-κB activity in VSMCs was observed by electrophoretic mobility shift assay. Results: The NF-κB activity of VSMCs was significantly increased after treatment with H2O2 or fetal bovine serum (NCS) for 72 h. However, the NF-κB activity was partially inhibited by adding 100 μmol / L probucol with the inhibitory rates of 37.1% And 14.8%. CONCLUSIONS: Probucol inhibits the activity of NF-κB induced by NCS and H2O2 in VSMCs, which may be one of the mechanisms for its effective prevention and treatment of RS and AS
