目的:从细胞分子水平探讨怀牛膝药物血清抗衰老作用及作用机理。方法:运用血清药理学方法,以体外培养的人胚肺二倍体成纤维细胞为材料,分为怀牛膝药物血清组、大鼠血清组和正常对照组,相同条件下传代培养,以生长曲线、MTT法、3H-TdR掺入法观察细胞增殖变化及DNA合成情况。结果:同一代龄,药物组细胞繁殖总数、OD490值、CPM掺入量等均明显高于对照组。结论:怀牛膝药物血清有较显著的促细胞增殖能力。
OBJECTIVE: To investigate the anti-aging effect and mechanism of anti-aging serum of Achyranthes bidentata from cell and molecular level. METHODS: Serum pharmacological methods were used to culture human embryonic lung diploid fibroblasts in vitro. The samples were divided into Huai-A-Knee drug serum group, rat serum group and normal control group. They were subcultured under the same conditions to grow. Curves, MTT assay, and 3H-TdR incorporation were used to observe cell proliferation and DNA synthesis. RESULTS: At the same age, the total cell population, OD490 value, and CPM incorporation in the drug group were significantly higher than those in the control group. Conclusion: The serum of Achyranthes bidentata has significant promoting cell proliferation.
