肥胖基因(ob)是近年刚被克隆的新基因,该基因产物Leptin是反映体内脂肪含量和调节体重的重要信号因子.首次报道猪ob基因全长序列,并对不同物种ob基因的同源性进行了比较.以 Uni朲APTM为载体构建了猪脂肪cDNA文库,根据已知的人和鼠ob基因序列设计PCR引物,利用PCR法筛选猪脂肪cDNA文库,并用RT-CR从脂肪RNA中扩增的366bp 猪ob基因片段作探针,获得了全长3 277bp 的猪ob基因cDNA的克隆和序列.ob基因序列在不同的物种之间具有很强的保守性.猪与人和鼠ob基因编码区核苷酸序列的同源性分别为88.5%和84.7%;猪ob基因编码的Leptin 蛋白氨基酸序列和人、鼠Leptin 蛋白的同源性分别为86.0%和83.4%.RT-CR分析表明:猪ob基因不仅在脂肪组织中大量表达,而且在肝脏、心脏、脾脏、肾脏、肌肉组织中也有微量表达.表明猪ob基因的表达调控与人和鼠可能存在差异.
