目的研究中药昆布水提物(thalluslami-nariaePE,TLPE)对耐药细胞株B-MD-C1(ADR+/+)耐药性的逆转作用。方法应用MTT法测定TLPE逆转多柔比星对B-MD-C(ADR+/+)的细胞毒作用,应用流式细胞仪(FCM)检测TLPE对多柔比星在细胞内的相对浓度和P-gp蛋白表达水平的影响。结果经TLPE作用后,多柔比星对B-MD-C1(ADR+/+)细胞的抑制率显著增高,P<0·01,明显提高了多柔比星进入并聚集于B-MD-C1(ADR+/+)细胞内的浓度,P<0·01;细胞的P-gp蛋白的表达明显降低,P<0·01;以上作用均与TLPE呈浓度依赖性。结论TLPE可提高多柔比星对耐药细胞的细胞毒作用,这种作用可能与抑制细胞P-gp的表达以及增加细胞内药物浓度有关。
Objective To study the reversal effect of thalluslami-nariaePE (TLPE) on the drug resistance of drug resistant cell line B-MD-C1 (ADR+/+). METHODS: MTT assay was used to determine the cytotoxicity of TLPE against doxorubicin against B-MD-C (ADR+/+). Flow cytometry (FCM) was used to determine the relative concentration of docetaxel in cells and the relative concentration of doxorubicin in cells. - The effect of gp protein expression levels. Results After TLPE treatment, the inhibition rate of doxorubicin on B-MD-C1 (ADR+/+) cells was significantly higher (P<0.01), which significantly increased the entrance of doxorubicin and accumulated in B-MD-C1. (ADR+/+) intracellular concentration, P<0.01; cell P-gp expression was significantly decreased, P<0.01; both effects were in a concentration-dependent manner with TLPE. Conclusion TLPE can increase the cytotoxicity of doxorubicin against drug-resistant cells. This effect may be related to inhibiting the expression of P-gp and increasing the intracellular drug concentration.
