研究了以2,4-二硝基氯苯(DNCB)作衍生化试剂在pH9.1的硼砂缓冲液中沸水浴加热1h,制得17种DNP-氨基酸,用于反相高效液相色谱(RP-HPLC)测定复方氨基酸的方法。采用C8柱,以乙腈-醋酸盐缓冲系统梯度洗脱,22min内17种氨基酸的DNP衍生物及衍生剂水解峰可较好分离;衍生化产物稳定,360nm处紫外检测,最低检出限为0.1μg。本法简便,重现性好,结果较为满意。
The effects of 2,4-dinitrochlorobenzene (DNCB) as derivatization reagent in boiling water bath at pH9.1 for 1 h were studied, and 17 kinds of DNP-amino acids were prepared for reversed-phase high performance liquid chromatography RP-HPLC) method for the determination of compound amino acids. Using C8 column, gradient elution with acetonitrile-acetate buffer system, the 17 amino acids of DNP derivative and derivatization peak can be separated well within 22 min. The derivatized product was stable and detected by UV at 360 nm with the detection limit of 0.1 μg. This method is simple, reproducible, the result is more satisfactory.
