目的研究罗布麻叶总黄酮提取物对慢性应激抑郁大鼠的抗抑郁作用,并探讨其对S100A10基因表达的影响。方法将105只SD大鼠随机分为7组,每组15只,根据给药成分与剂量分为对照组、抑郁组,低、高剂量预防组(分别给予罗布麻叶总黄酮提取物40、80 mg/kg),低、高剂量治疗组(分别给予罗布麻叶总黄酮提取物40、80 mg/kg)和氟西汀组(氟西汀20 mg/kg)。采用慢性不可预知性轻度应激(CUMS)法建立大鼠抑郁模型,通过旷场实验观察各组大鼠垂直运动次数及水平运动总路程,采用实时荧光定量PCR方法检测大鼠海马区S100A10 mRNA表达。结果与对照组相比,抑郁组、罗布麻叶总黄酮提取物各干预组和氟西汀组大鼠在应激4周后旷场实验中的水平运动总路程及垂直活动次数减少(P<0.05);与抑郁组相比,罗布麻叶总黄酮提取物高预防组和高治疗组的垂直运动次数,及罗布麻叶总黄酮提取物各干预组的水平运动总路程显著增加(P<0.05);氟西汀组垂直运动次数及水平运动总路程比抑郁组增加(P<0.05)。与对照组相比,抑郁组大鼠海马组织S100A10基因表达水平升高,罗布麻叶总黄酮提取物低预防组和低治疗组S100A10基因表达水平均降低(P<0.05);与抑郁组相比,罗布麻叶总黄酮提取物各干预组和氟西汀组S100A10基因表达水平均降低(P<0.05)。结论罗布麻叶总黄酮提取物对CUMS大鼠具有抗抑郁样行为作用,调节S100A10基因表达可能是罗布麻叶总黄酮提取物抗抑郁作用机制之一。
Objective To study the antidepressant effect of Apocynum venetum flavonoids extract on chronic stress-induced depression in rats and its effect on S100A10 gene expression. Methods 105 Sprague-Dawley rats were randomly divided into 7 groups (15 rats in each group). The rats were divided into control group, depression group, low-dose and high-dose prevention group 80 mg / kg), low and high dose treatment groups (40, 80 mg / kg) and fluoxetine (fluoxetine 20 mg / kg). The chronic depression model was established by using CUMS method. The vertical movement frequency and the total distance of horizontal movement were observed in open-field test. The expression of S100A10 mRNA in hippocampus of rats was detected by real-time fluorescence quantitative PCR expression. Results Compared with the control group, the total distance of horizontal movement and the number of vertical movements in the depression group, the total flavonoids extract of Apocynum venetum leaves group and the fluoxetine group rats in the open field test after 4 weeks of stress decreased (P < 0.05). Compared with the depression group, the total number of vertical movement of the total flavonoids extract of the Apocynum venetum extract group and the high treatment group and the total exercise of the total flavonoid extract of the Apocynum venetum leaves extract group increased significantly (P <0.05 ); Fluoxetine group the number of vertical movement and horizontal exercise total distance than the depression group increased (P <0.05). Compared with the control group, the expression level of S100A10 gene in hippocampus of depression group was increased, the expression level of S100A10 gene in Apocynum venetum extract low-prevention group and low-treatment group were decreased (P <0.05); Compared with depression group , The total flavonoids extract of Apocynum venetum extract intervention group and fluoxetine group S100A10 gene expression levels were reduced (P <0.05). Conclusion The total flavonoids extract of Apocynum venetum leaves has antidepressant-like behavior in CUMS rats. Regulating S100A10 gene expression may be one of the mechanisms of anti-depressant effect of Apocynum venetum flavonoids extract.
