以药用植物鹅毛玉凤花的种子胚为外植体进行培养,研究不同无机盐浓度、活性炭浓度、NAA浓度、碳源、pH值及光照时间等因子对其胚萌发的影响。结果表明:以固体1/2 MS为基本培养基,添加NAA 0.5 mg/L+3%蔗糖和0.1%~0.5%活性炭,培养基pH为5.5~6.0时对种胚的萌发较好;先黑暗培养再光照条件培养可以提高胚的萌发率,达82%。
The seed embryo of medicinal plant Eupatorium mandshurica was used as explants to study the effects of different inorganic salt concentration, activated carbon concentration, NAA concentration, carbon source, pH value and light time on embryo germination. The results showed that the seed germination was better when 1/2 MS solid medium was added with NAA 0.5 mg / L + 3% sucrose and 0.1% ~ 0.5% activated carbon at pH 6.0-6.0. Culture and then light conditions can improve embryo germination rate, up to 82%.
