目的通过建立2次打击大鼠致多器官功能障碍综合征(MODS)的动物模型,探讨黄芪对不同复苏液体复苏后MODS中血管内皮细胞损伤的影响。方法将32只SD大鼠随机分为对照组、乳酸林格液复苏组(RL组)、7.5%的高渗盐水和6%的羟乙基淀粉混合液复苏组(HTH组),HTH组中分为复苏后未注射黄芪组(HTH1组)和复苏后注射黄芪组(HTH2组)。RL组和HTH组复制大鼠MODS复苏的动物模型,HTH2组腹腔注入黄芪注射液,HTH1组和RL组注入0.9%氯化钠溶液。分别在注射黄芪注射液和0.9%氯化钠溶液后24 h采集血液和肺组织标本,测定大鼠血清中循环内皮细胞(CEC)数量、一氧化氮(NO)、内皮素-1(ET-1)、肺组织中肿瘤坏死因子-α(TNF-α)和细胞间黏附分子(ICAM-1)的含量、白细胞介素-6(IL-6)、血管细胞黏附分子(VCAM-1)mRNA,肺组织标本行光镜下病理检查。结果 HTH2组大鼠腹腔注射黄芪注射液后24 h CEC数量、NO、ET-1含量明显低于RL组、HTH1组(均为P<0.05);HTH2组大鼠腹腔注射黄芪注射液后24 h TNF-α、ICAM-1、IL-6、VCAM-1含量明显低于HTH1组(均为P<0.05)。结论黄芪注射液对不同复苏液体复苏后MODS中血管内皮细胞有明显的保护作用。
OBJECTIVE: To establish two animal models of multiple organ dysfunction syndrome (MODS) in rats and investigate the effect of Radix Astragali on the injury of vascular endothelial cells in MODS after resuscitation with different resuscitation fluids. Methods 32 SD rats were randomly divided into control group, lactated Ringer ’s solution group (RL group), 7.5% hypertonic saline group and 6% hydroxyethyl starch mixture recovery group (HTH group), HTH group Divided into astragalus group (HTH1 group) and astragalus group (HTH2 group) after resuscitation. In the RL group and HTH group, the animal model of MODS resuscitation was established in rats. HTH2 group was given intraperitoneal injection of astragalus injection, and HTH1 group and RL group were injected with 0.9% sodium chloride solution. The blood and lung tissue samples were taken 24 hours after injection of astragalus injection and 0.9% sodium chloride solution respectively. The levels of circulating endothelial cells (CEC), nitric oxide (NO) and endothelin-1 (ET- 1), the content of TNF-α and ICAM-1, the expression of interleukin-6 (IL-6) and vascular cell adhesion molecule (VCAM- , Lung tissue specimens under light microscope pathological examination. Results The number of CEC, the contents of NO and ET-1 in HTH2 group were significantly lower than those in RL group and HTH1 group (all P <0.05) at 24 h after intraperitoneal injection of Astragalus injection. In HTH2 group, The levels of TNF-α, ICAM-1, IL-6 and VCAM-1 were significantly lower than those of HTH1 group (all P <0.05). Conclusion Astragalus injection has obvious protective effect on vascular endothelial cells in MODS after resuscitation with different resuscitation fluids.
