目的:在体外实验中,探讨血小板和红细胞对T细胞亚群的调控作用。方法:流式细胞术测定血小板CD59、红细胞CD59、T细胞CD3+CD4+、CD3+CD8+的表达水平。按不同的实验条件进行分组,全血细胞组、有PL无红细胞(RBC)组、无血小板(PL)组、无RBC无PL组。同时观测各组抗原刺激前后T细胞变化。结果:各组T细胞CD3+CD4+、CD3+CD4+/CD3+比较:全血细胞组>有PL无RBC组,抗原刺激前后(P<0.001);有PL无RBC组>无RBC无PL组刺激前(P>0.05)刺激后(P<0.05)。各组T细胞CD3+CD8+表达无明显变化。各组CD3+CD8+/CD3+比较:全血细胞组<有PL无RBC组抗原刺激前后(P<0.01);有PL无RBC组<无RBC无PL组刺激前(P>0.05)刺激后(P<0.05)。全血细胞组和无PL组比较各项指标均无统计学意义(P>0.05)。抗原刺激前RBCCD59与CD3+CD4+在全血细胞组中负相关(P<0.05);而在无PL组中则无相关性;PLCD59与CD3+CD8+CD3+全血细胞组中的正相关无统计学意义(P>0.05);而在无红细胞组中则呈负相关(P<0.05)。结论:活化血小板、红细胞均可正向调节CD3+CD4+/CD3+,负向调节CD3+CD8+/CD3+。
Objective: To investigate the regulatory effect of platelets and erythrocytes on T cell subsets in vitro. Methods: Flow cytometry was used to detect the expression of platelet CD59, erythrocyte CD59, T cells CD3 + CD4 +, CD3 + CD8 +. According to different experimental conditions grouping, whole blood cells group, PL non-erythrocyte (RBC) group, no platelet (PL) group, no RBC PL group. At the same time, the changes of T cells before and after antigen stimulation were observed. Results: The comparison of CD3 + CD4 + and CD3 + CD4 + / CD3 + of T cells in all groups: PL group without RBC, before and after antigen stimulation (P <0.001); PL without RBC group> P> 0.05) after stimulation (P <0.05). There was no significant change of CD3 + CD8 + expression in each group of T cells. The levels of CD3 + CD8 + / CD3 + in the whole blood group before and after stimulation with PL without RBC antigen (P <0.01) 0.05). There was no significant difference between the whole blood group and the non-PL group (P> 0.05). There was a negative correlation between RBCCD59 and CD3 + CD4 + in whole blood cells before antigen stimulation (P <0.05), but no correlation between PLD and PLCD59. There was no significant correlation between PLCD59 and CD3 + CD8 + CD3 + whole blood cells (P> 0.05), but not in erythrocyte group (P <0.05). Conclusion: Both activated platelets and erythrocytes can positively regulate CD3 + CD4 + / CD3 + and negatively regulate CD3 + CD8 + / CD3 +.
