目的探讨COX-2(cyclooxygenase-2)蛋白在鼻咽癌组织中的表达及其临床意义。方法分别应用免疫组化方法(IHC)和蛋白印迹方法(Western Blot)检测47例鼻咽癌组织(其中用IHC33例,Western Blot14例)及24例鼻咽部炎症组织中COX-2蛋白的表达。结果COX-2蛋白在鼻咽癌组织的阳性表达率为81.8%(27/33),在鼻咽炎组织的阳性表达率为25%(3/12),两者表达差异有统计学意义(P=0.001)。Western Blot结果显示,在两种组织中COX-2蛋白表达的相对值分别为1.258±0.034和0.487±0.076,在鼻咽癌组表达显著增高(P=0.000)。结论COX-2蛋白的高度表达在鼻咽癌的发生、发展中起重要作用,选择性COX-2抑制剂有望成为鼻咽癌化疗的重要药物。
Objective To investigate the expression of cyclooxygenase-2 (COX-2) in nasopharyngeal carcinoma and its clinical significance. Methods Immunohistochemistry (IHC) and Western Blot were used to detect the expression of COX-2 protein in 47 cases of nasopharyngeal carcinoma (IHC) and Western Blot (14 cases of IHC), and 14 cases of nasopharyngeal inflammation . Results The positive expression rate of COX-2 protein in nasopharyngeal carcinoma was 81.8% (27/33) and that in nasopharyngeal tissues was 25% (3/12), the difference was statistically significant (P = 0.001). The results of Western Blot showed that the relative expression of COX-2 protein in the two tissues were 1.258 ± 0.034 and 0.487 ± 0.076, respectively, which were significantly increased in NPC tissues (P = 0.000). Conclusion The high expression of COX-2 protein plays an important role in the occurrence and development of nasopharyngeal carcinoma. Selective COX-2 inhibitor is expected to become an important drug for the treatment of nasopharyngeal carcinoma.
