目的探讨丹瓜方对Apo E^-/-糖尿病模型小鼠脑组织胰岛素样生长因子-1(insulin-like growth factor-1,IGF-1)表达水平的影响,以探究丹瓜方防治糖尿病脑病的部分机制。方法以链脲佐菌素(STZ)对Apo E^-/-小鼠塑造糖尿病模型。将成模后的小鼠按体质量分层,依照随机数字表随机分为模型组、丹瓜方组、吡格列酮组、联合组,每组8只。选取血糖正常的同龄同品系C57小鼠8只作为对照组。分组灌胃干预12周。ELISA法测定IGF-1表达。结果与C57组IGF-1(19.89±0.95)ng/m L比较,其余各组均明显降低(P<0.01);丹瓜方组(14.31±0.99)ng/m L和联合用药组IGF-1水平(11.58±0.92)ng/m L明显高于模型组(7.25±0.42)ng/m L;丹瓜方组IGF-1水平明显高于联合用药组(P<0.01)。结论通过适度提升糖尿病鼠脑组织中IGF-1表达,可能是丹瓜方防治糖尿病脑病的部分机制。
Objective To investigate the effect of Danggu recipe on the expression of insulin-like growth factor-1 (IGF-1) in brain tissue of Apo E ^ - / - diabetic mice and explore the mechanism of Danggua recipe in preventing and treating diabetic encephalopathy Part of the mechanism. Methods Streptozotocin (STZ) was used to model diabetes mellitus in Apo E ^ - / - mice. The mice were divided into body mass according to the random number table randomly divided into model group, Dan Gua group, pioglitazone group, the combination group, each group of eight. Eight normal-aged C57 mice of the same age with normal blood glucose were selected as the control group. Grouping gavage intervention for 12 weeks. ELISA method for the determination of IGF-1 expression. Results Compared with IGF-1 (19.89 ± 0.95) ng / m L in C57 group, the other groups were significantly decreased (P <0.01) (11.58 ± 0.92) ng / m L was significantly higher than that of model group (7.25 ± 0.42) ng / m L; IGF-1 level in Dangaifan group was significantly higher than that in combination group (P <0.01). Conclusion It is possible that Danggua recipe can prevent and treat diabetic encephalopathy through moderately increasing IGF-1 expression in diabetic rat brain.
