目的建立快速酸浸取石墨炉原子吸收法测定全血微量铅、镉、铬的方法。方法血样加5%硝酸后采用振荡、高速离心法进行样品前处理,以2%磷酸二氢铵与0.2%硝酸镁混合液为基改剂,塞曼扣背景及标准加入法对样品中的微量铅、镉、铬进行测定。结果铅、镉和铬分别在5μg/L~60μg/L、0.5μg/L~6μg/L和2μg/L~20μg/L时具有良好的线性,相关系数均>0.999,RSD为2.31%~5.29%,回收率为95.3%~104.0%。采用本法对美国Wisconsin State Laboratory of Hygiene全球血铅比对样品进行方法验证,所测结果均在标准参考值内。结论本法具有快速、简便、环保、准确的优点,适合批量同时分析全血中铅、镉、铬。
Objective To establish a rapid acid leaching method for the determination of trace lead, cadmium and chromium in whole blood by graphite furnace atomic absorption spectrometry. Methods The blood samples were treated with 5% nitric acid by shaking and high speed centrifugation. The samples were pretreated with 2% ammonium dihydrogen phosphate and 0.2% magnesium nitrate mixture. Zeeman buckling background and standard addition method were used to determine the trace amount Lead, cadmium, chromium were measured. Results Lead, cadmium and chromium showed good linearity at 5μg / L ~ 60μg / L, 0.5μg / L ~ 6μg / L and 2μg / L ~ 20μg / L respectively with correlation coefficients> 0.999 and RSDs ranging from 2.31% to 5.29 %, The recovery rate was 95.3% ~ 104.0%. This method is used to validate the method of global blood lead ratio of Wisconsin State Laboratory of Hygiene in the United States. The measured results are within the standard reference values. Conclusion This method is fast, simple, environmentally friendly and accurate. It is suitable for simultaneous and batch analysis of lead, cadmium and chromium in whole blood.
