目的 研究切割 bcl- 2 m RNA核酶诱导人胆管癌细胞凋亡的形态学改变 .方法 将合成的切割 bcl- 2 m RNA核酶基因与真核细胞表达载体重组 ,并用重组后的载体转染人胆管癌细胞 ,通过光镜及扫描和透射电镜手段 ,观察转染后的胆管癌细胞凋亡的形态学改变 .结果 转染切割 bcl- 2 m R-NA核酶基因的胆管癌细胞出现较典型的凋亡形态学改变 .结论 该切割 bcl- 2 m RNA核酶能明显降低胆管癌细胞内bcl- 2的表达水平并能够诱导其出现自发性凋亡
OBJECTIVE: To study the morphological changes induced by cleavage of bcl-2 mRNA in human cholangiocarcinoma cells.Methods The recombinant bcl-2 m RNA ribozyme gene was recombined with the eukaryotic cell expression vector and transfected with the recombinant vector Human cholangiocarcinoma cells were observed by light microscopy and scanning electron microscopy and transmission electron microscopy to observe the morphological changes of apoptotic cells.Results The bcl-2 mRNA transfected cholangiocarcinoma cells transfected with bcl- Typical apoptotic morphological changes.Conclusion The cleavage of bcl-2 m RNA ribozyme can significantly reduce bcl-2 expression in cholangiocarcinoma cells and induce spontaneous apoptosis
