目的 了解APOE基因多态性在中老年人群中的分布.方法 采用多聚酶链反应限制性片段长度多态性技术(PCR-RFLP),检测广州市荔湾区512 名年龄55 岁及以上的健康志愿者的APOE基因型,特异性扩增APOE基因第四外显子区含编码112 位至158 位氨基酸的基因序列,产物片段244 BP,用限制性内切酶HHAI酶解PCR产物,应用聚丙烯酰胺凝胶电泳进行酶解产物的基因分型.结果 广州市荔湾区健康中老年人群中,APOE等位基因频率分别为ε2∶9.2% ,ε3∶84.2% ,ε4∶6.6% ,ε2 频率随年龄增加有上升趋势,其中65~74 岁年龄组与55~64 岁年龄组比较,差异有显著性意义(P< 0.05),ε4 频率则随年龄增加而明显下降(P< 0.05).APOE等位基因频率分布与男女性别无关.结论 广州市荔湾区健康中老年人群中的APOE等位基因频率的分布与国内其他多数健康人群大体相同,与国外同类研究相比则有差异.“,”To study the distribution of genetic polymorphism of ApoE in the mid aged and elderly population.Methods Polymerase chain reaction(PCR) and restriction fragment length polymorphism(RFLP) were used to determine ApoE genotypes for 512 healthy individuals (ages≥55 years) in Liwang district of Guangzhou. ApoE gene sequences coded for amino acid 112 to 158, 244 bp in length were amplified specificly. The PCR productions were digested by Hha I. The genotypes of digested productions were analysed by polyacrylamide nondenaturing gel.Results The results showed the frequencies of ApoE ε2, ε3 and ε4 alleles, were 9.2%, 84.2%, and 6.6% respectively. The frequency of ApoE ε2 allele in the elderly group(65~74 years) was increased obviously (P<0 05) as compared with that of the mid aged group(55~64 years). The frequency of ApoE ε4 allele was decreased obviously (P<0 05) . No gender difference on the frequency distribution of ApoE allele were observed.Conclusions The distribution of genetic polymorphism of ApoE in the mid aged and elderly population in Liwang district of Guangzhou is similar to that of most domestic healthy population but different to that in foreign studies.
