目的了解急性心肌梗死(acute myocardial infarction,AMI)后心室肌HCN2、HCN4的mRNA和蛋白质表达的变化趋势,探讨普伐他汀对AMI后心室肌HCN2、HCN4变化的影响。方法通过结扎大鼠左冠状动脉前降支建立AMI模型,将存活的20只大鼠按随机数字表法分为24h组、1周组、4周组和普伐他汀组,每组5只,同时于各时间点均设立假手术组,每组5只。取右室心肌组织样本,用Real-timePCR检测HCN2、HCN4 mRNA的表达,用Western blot检测HCN2、HCN4蛋白质表达。结果4周组和普伐他汀组HCN2 mRNA及蛋白质表达,HCN4 mRNA表达均高于假手术组(P<0.05);普伐他汀组HCN2 mRNA及蛋白质表达明显低于4周组(P<0.05)。各组均未检测到HCN4蛋白质表达。结论AMI后4周心室肌HCN2、HCN4表达增高;普伐他汀能抑制AMI后HCN2表达的上调,而对HCN4无明显影响。
Objective To investigate the changes of mRNA and protein expression of HCN2 and HCN4 in acute myocardial infarction (AMI) and to investigate the effect of pravastatin on changes of HCN2 and HCN4 in ventricular myocytes after acute myocardial infarction (AMI). Methods AMI model was established by ligation of left anterior descending coronary artery in rats. Twenty rats survived were divided into 24h group, 1 week group, 4 weeks group and pravastatin group by random number table, At the same time, sham-operated group was established at each time point, 5 in each group. Right ventricular myocardial tissue samples were taken and the expression of HCN2 and HCN4 mRNA was detected by Real-time PCR. HCN2 and HCN4 protein expression was detected by Western blot. Results HCN2 mRNA and protein expression and HCN4 mRNA expression in 4-week group and pravastatin group were significantly higher than those in sham-operated group (P <0.05). HCN2 mRNA and protein expression in pravastatin group was significantly lower than that in 4-week group (P <0.05) . HCN4 protein expression was not detected in all groups. Conclusion The expression of HCN2 and HCN4 in ventricular myocardium increased 4 weeks after AMI. Pravastatin could inhibit the up-regulation of HCN2 expression after AMI, but had no effect on HCN4.
