[目的]探讨中波紫外线(UVB)对正常小鼠表皮细胞凋亡及凋亡相关基因Bax和Bcl-2的调控。[方法]将正常ICR小鼠随机分为对照组(假照射组)、500J/m2、1000J/m2、1500J/m2、2000J/m2紫外线照射后2h组(每组10只小鼠)。1500J/m2紫外线照射后4h组、8h组、12h组、24h组、48h组、72h组(每组10只小鼠)。采用链霉菌亲生物素蛋白-过氧化物酶标免疫组织化学法(streptavidin-peroxidase,SP)染色检测凋亡相关基因Bax和Bcl-2的蛋白表达。[结果]小鼠皮肤用不同剂量紫外线照射2h后Bax的表达,随紫外线剂量增大而变大,Bcl-2表达减小,呈剂量依赖性。用1500J/m2紫外线照射小鼠后12hBax表达达到高峰,Bcl-2表达减弱;24h后Bax表达开始减弱,而Bcl-2则开始表达,72h恢复正常,Bax和Bcl-2的表达与紫外线照射后时间有关。[结论]中波紫外线照射小鼠皮肤后,Bax表达上调,Bcl-2表达下调,促进小鼠紫外线损伤的表皮细胞凋亡。
[Objective] To investigate the effects of ultraviolet B (UVB) on the apoptosis of epidermic cells and the regulation of apoptosis-related genes Bax and Bcl-2 in normal mice. [Methods] The normal ICR mice were randomly divided into control group (sham irradiation group), 500J / m2, 1000J / m2, 1500J / m2 and 2000J / m2 for 2h after UV irradiation (10 mice per group). 4h group, 8h group, 12h group, 24h group, 48h group and 72h group (10 mice in each group) after 1500J / m2 UV irradiation. Streptavidin-peroxidase (SP) staining was used to detect the protein expression of apoptosis-related genes Bax and Bcl-2 by Streptomyces avidin-streptavidin-peroxidase (SP) staining. [Result] The expression of Bax in mouse skin irradiated with different doses of UV light for 2h increased with the increase of UV dose, and the expression of Bcl-2 decreased in a dose-dependent manner. The expression of Bcl-2 was up-regulated at 12 hours after irradiation with 1500J / m2 ultraviolet light. The expression of Bcl-2 was decreased after 24 hours, while the expression of Bcl-2 began to return to normal after 72 hours. The expression of Bax and Bcl- Time related. [Conclusion] After UVB irradiation, the expression of Bax and Bcl-2 were up-regulated in the skin of mice, which promoted the apoptosis of the epidermis cells in UV-exposed mice.
