目的探讨人乳腺癌中原癌基因c-myc与hTERT mRNA表达的关系。方法收集癌旁正常乳腺组织、导管原位癌、浸润性导管癌各12、18和25例,用原位杂交法检测hTERT mRNA表达,并用免疫组织化学法检测乳腺导管癌c-myc表达。结果①hTERT mRNA在癌旁正常乳腺组织中未见表达,在导管原位癌、浸润癌中的阳性率分别为83.33%和88.00%,后两组hTERT mRNA表达明显高于正常组;②hTERT mRNA表达与浸润性导管癌肿块大小及淋巴结转移与否无相关性(P>0.05);③43例乳腺导管癌中hTERT mRNA与c-myc表达呈正相关(γ=0.3884,P<0.05)。结论端粒酶hTERT的表达可能在乳腺导管癌的组织发生中起关键作用,c-myc的过度表达可能与乳腺导管癌hTERT基因转录激活有关。
Objective To investigate the relationship between c-myc and hTERT mRNA expression in human breast cancer. Methods The specimens of normal breast tissues, ductal carcinoma in situ and invasive ductal carcinoma were collected from 12, 18 and 25 cases respectively. The expression of hTERT mRNA was detected by in situ hybridization and the expression of c-myc was detected by immunohistochemistry. Results ① The expression of hTERT mRNA was not detected in adjacent normal breast tissue, and the positive rates of hTERT mRNA in ductal carcinoma and invasive carcinoma were 83.33% and 88.00% respectively. The expression of hTERT mRNA in the latter two groups was significantly higher than that in normal group. There was no correlation between tumor size and lymph node metastasis in infiltrating ductal carcinoma (P> 0.05). (3) There was a positive correlation between the expression of hTERT mRNA and c-myc in 43 cases of breast ductal carcinoma (r = 0.3884, P <0.05). Conclusion The expression of telomerase hTERT may play a key role in the carcinogenesis of breast ductal carcinoma. The overexpression of c-myc may be related to the transcriptional activation of hTERT gene in breast ductal carcinoma.
