The endoplasmic reticulum in the whole-mount cultured cellsfixed with our new Prescription of potassium permanganate andexamined by standard transmission electron microscopy at 60KV.The present potassium permanganate fixative destroys proteinaceousstructures but preserve the membrane lipids in the membranousorganelles of the cells, hence the endoplasmic reticulum can beobserved as a net-like membranous system composed of tubularcanaliculi and f1 attened cisternae. The tubular canaliculi have adiameter of 30-40nm and the flattened cisternae possess irregularspaces of 50~80nm. Using this method, the real distribution andpattern formation of the endoplasmie reticulum in wholemountcultured cells can be clearly observed, since membranes are sharplydefined from the surrounding cytoplasm.
