目的探讨转化生长因子β(TGF-β)在阿司匹林抑制细胞增殖中的作用。方法选用大鼠胸椎动脉血管平滑肌细胞检测阿司匹林的细胞毒性及其对细胞增殖和细胞周期的影响。结果阿司匹林对血管平滑肌细胞具有抑制作用,且在第6天达到最强,而抗TGF-β抗体可逆转阿司匹林对细胞增殖的抑制。结论 TGF-β可介导阿司匹林在细胞增殖中的抑制作用。因此推断,阿司匹林预防冠状动脉疾病中的机制并不仅限于抗血小板作用,还可能依赖于其抑制斑块生长的作用。
Objective To investigate the role of transforming growth factor β (TGF-β) in the inhibition of cell proliferation by aspirin. Methods Rat thoracic artery smooth muscle cells were used to detect the cytotoxicity of aspirin and its effect on cell proliferation and cell cycle. Results Aspirin had an inhibitory effect on vascular smooth muscle cells and reached the strongest on day 6, whereas anti-TGF-β antibody reversed aspirin’s inhibition of cell proliferation. Conclusion TGF-β can mediate the inhibitory effect of aspirin on cell proliferation. Therefore, it is inferred that the mechanism of aspirin’s prevention of coronary artery disease is not limited to the anti-platelet effect but may depend on its role in inhibiting plaque growth.
