目的探讨弓形虫感染对小鼠血睾屏障及生精过程的影响。方法选取9~10周龄雄性BALB/C小鼠45只,随机分为正常对照组和弓形虫速殖子感染低剂量组及高剂量组两感染组,分别腹腔注射1×102和1×103个弓形虫速殖子/只。感染后第6d,ELISA检测血清IFN-γ及抗精子抗体(AsAb)水平,硝酸还原法测定睾丸组织一氧化氮(NO)含量,病理切片及硝酸镧示踪透射电镜观察血睾屏障和生精细胞损伤情况。结果正常组小鼠血清IFN-γ维持在低水平[(32.88±5.68)pg/ml],低、高剂量组分别升高至(105.03±11.80)pg/ml和(312.61±44.86)pg/ml,差异具有统计学意义(P<0.01);同时低剂量组[(37.29±4.40)μmol/L]和高剂量组[(40.13±3.25)μmol/L]睾丸NO水平也较正常组[(29.48±4.22)μmol/L]显著增高(P<0.01)。感染组小鼠睾丸组织结构混乱、生精细胞减少,电镜观察显示低剂量组硝酸镧渗入血睾屏障,生精细胞结构尚清晰;高剂量组血睾屏障隔离功能降低并伴有细胞凋亡。正常组小鼠血清AsAb为(50.49±14.59)UI/ml,低、高剂量组分别为(105.28±34.90)UI/ml和(193.55±74.8)UI/ml,差异有统计学意义(P<0.01)。结论弓形虫感染导致小鼠血睾屏障损伤,可能与NO、IFN-γ等调节因子异常有关,进而引起AsAb升高及生精细胞损伤,导致生精障碍。
Objective To investigate the effect of Toxoplasma gondii infection on the blood-testis barrier and spermatogenesis in mice. Methods Forty-five male BALB / C mice aged 9-10 weeks were randomly divided into two groups: normal control group, tachyzoite-fed tachyzoites and high-dose tachyzoites. The mice were injected intraperitoneally with 1 × 102 and 1 × 103 Toxoplasma tachyzoites / only. Serum levels of IFN-γ and anti-sperm antibody (AsAb) were detected by ELISA 6 days after infection. Nitric oxide (NO) content of testis was determined by nitrate reduction method. Pathological examination and lanthanum nitrate tracing transmission electron microscope Cell damage. Results The serum levels of IFN-γ in normal mice were maintained at low level [(32.88 ± 5.68) pg / ml], while those in low and high dose groups were increased to (105.03 ± 11.80) pg / ml and (312.61 ± 44.86) pg / ml (P <0.01), while the levels of NO in low dose group [(37.29 ± 4.40) μmol / L] and high dose group [(40.13 ± 3.25) μmol / L] ± 4.22) μmol / L] (P <0.01). In the infected mice, the structure of testis was disorganized and the number of spermatogenic cells decreased. Electron microscopy showed lanthanum nitrate infiltrated into the blood testis barrier in low dose group, and the structure of spermatogenic cells was still clear. The isolation of blood and testis barrier in high dose group was accompanied with apoptosis. The serum AsAb of normal mice was (50.49 ± 14.59) UI / ml, while the low and high doses of AsAb were (105.28 ± 34.90) UI / ml and (193.55 ± 74.8) UI / ml respectively ). Conclusion Toxoplasma gondii infection can lead to damage of blood-testis barrier in mice, which may be related to the abnormal regulation of NO and IFN-γ and other factors, leading to the increase of AsAb and the damage of spermatogenic cells.
