本文通过羟胺将天花粉蛋白(TCS)进行特异性切割产生的大、小二个片段(HATf1、HATf2),用SDSPAGE对其表观分子量进行测定;应用免疫印迹法研究了其抗原性的分布,根据反应格局的不同,选取了第14号、第16号单抗作为探针,在亚细胞水平研究了它们对TCS的封闭作用。结果发现,第14号单抗具有封闭效应而第16号则无,结合免疫印迹的格局并从空间构象上可以推测天花粉蛋白的活性中心位于大、小片段交界处的大片段上。
In this paper, the large and small two fragments (HATf1, HATf2) that were produced by the specific cleavage of Trichosanthin (TCS) by hydroxylamine were determined and their apparent molecular weights were determined by SDSPAGE. The antigenicity distribution was studied by Western blotting, According to the different reaction patterns, the 14th and 16th mAbs were selected as probes to study their blocking effects on TCS at the subcellular level. As a result, it was found that No. 14 McAb had a blocking effect and No. 16 did not. Combined with the pattern of the immunoblot, it was speculated from the spatial conformation that the active center of Trichosanthin protein is located on a large fragment at the juncture of large and small fragments.
