目的:观察FA-MNP-MMP-9-ASODN纳米复合物对鼻咽癌HNE-1细胞的体外抑制效应。方法:采用RT-PCR、Western-blot方法、MTT方法、流式细胞技术及Matrigel体外侵袭实验观察转染FA-MNP-MMP-9-ASODN 48h后HNE-1细胞MMP-9mRNA及蛋白表达水平和细胞增殖能力、凋亡和侵袭能力的变化。结果:FA-MNP-MMP-9-ASODN组HNE-1细胞MMP-9 mRNA和蛋白的表达水平较对照组及FA-MNP-MMP-9-NSODN无义序列组明显降低。同时FA-MNP-MMP-9-ASODN组HNE-1细胞生长抑制率约为35.66%,增殖活性较对照组和无义序列组明显降低。且HNE-1细胞周期也受到抑制,细胞凋亡率约为12.60%,明显高于对照组和无义序列组。侵袭实验显示FA-MNP-MMP-9-ASODN组穿膜细胞数约为21.00个,明显低于对照组和无义序列组。结论:FA-MNP-MMP-9-ASODN纳米复合物通过抑制MMP-9的表达,可降低鼻咽癌细胞的增殖和侵袭能力,促进凋亡,体外抑制效果良好。
AIM: To investigate the inhibitory effect of FA-MNP-MMP-9-ASODN nanocomposites in vitro on nasopharyngeal carcinoma HNE-1 cells. Methods: The mRNA and protein expression of MMP-9 in HNE-1 cells transfected with FA-MNP-MMP-9-ASODN for 48 hours were determined by RT-PCR, Western-blot, MTT, flow cytometry and Matrigel in vitro invasion assay Cell Proliferation, Apoptosis and Invasion. Results: The expression of MMP-9 mRNA and protein in HNE-1 cells in FA-MNP-MMP-9-ASODN group was significantly lower than that in control group and FA-MNP-MMP-9-NSODN nonsense sequence group. At the same time, the growth inhibition rate of HNE-1 cells in FA-MNP-MMP-9-ASODN group was 35.66%, and the proliferation activity was significantly lower than that in control group and non-sense sequence group. And HNE-1 cell cycle was also inhibited, the rate of apoptosis was about 12.60%, significantly higher than the control group and nonsense sequence group. Invasion experiments showed that the number of transmembrane cells in FA-MNP-MMP-9-ASODN group was about 21.00, which was significantly lower than that in control group and non-sense sequence group. CONCLUSION: FA-MNP-MMP-9-ASODN nanocomposites can inhibit the proliferation and invasion of nasopharyngeal carcinoma cells by inhibiting the expression of MMP-9 and promote apoptosis. The inhibitory effect is good in vitro.
