目的探讨七叶灵颗粒在抑制肿瘤生长及肿瘤细胞侵袭和转移方面的作用机制。方法将接种的C57小鼠随机分为4组:不用任何治疗的模型组(A组)、化疗组(B组)、七叶灵颗粒组(C组)和化疗加七叶灵颗粒组(D组),观察各组小鼠生存质量,比较各组小鼠的体内抑瘤作用。采用原位细胞凋亡检测试剂盒检测标本组织凋亡细胞数,用原位杂交检测,检测标本组织MMP9阳性表达数。结果①C组和D组小鼠的生存质量优于A组和B组。②D组的抑瘤率为74.47%,显著高于A组(P<0.01)。B、C、D组的平均瘤重显著低于A组(P均<0.05)。③原位细胞凋亡检测表明,B组、C组和D组瘤组织凋亡细胞数均显著高于A组(P均<0.05),且D组显著高于C组(P<0.05)。④原位杂交检测表明,D组、C组和B组肺癌组织中的MMP9表达水平均显著低于A组(P均<0.05)。结论七叶灵颗粒有一定的改善C57荷瘤小鼠生存质量、抑制Lewis肺癌细胞生长、诱导Lewis肺癌细胞凋亡及抑制MMP9活性表达的作用。
Objective To investigate the mechanism of Qiyeling granule in inhibiting tumor growth and tumor cell invasion and metastasis. Methods C57 mice were randomly divided into 4 groups: untreated model group (group A), chemotherapy group (group B), Qiyeling granules group (group C) and chemotherapy plus Qiyeling granules group (D Group), observed the quality of life of mice in each group, compared the inhibition effect of each group of mice in vivo. The number of apoptotic cells was detected by in situ apoptosis detection kit. The number of MMP9 positive cells was detected by in situ hybridization. Results ① The quality of life of group C and group D was better than group A and group B. ② The tumor inhibition rate of group D was 74.47%, which was significantly higher than that of group A (P <0.01). The average tumor weight in B, C and D groups was significantly lower than that in A group (all P <0.05). ③ In situ cell apoptosis assay showed that the number of apoptotic cells in group B, group C and group D were significantly higher than that in group A (P <0.05), and group D was significantly higher than group C (P <0.05). ④ In situ hybridization test showed that the expression levels of MMP9 in group D, group C and group B were significantly lower than those in group A (all P <0.05). Conclusion Qiyeling Granules can improve the quality of life of C57-bearing mice, inhibit the growth of Lewis lung cancer cells, induce the apoptosis of Lewis lung cancer cells and inhibit the expression of MMP9.
