目的:研究大孔树脂吸附纯化黄精小分子糖的最佳工艺条件及参数。方法:以小分子糖保留率和纯度为考察指标,比较四种大孔树脂对黄精小分子糖的吸附性能,考察上样浓度、上样量、洗脱流速和洗脱剂用量等因素对优选树脂纯化工艺的影响,确定最佳工艺参数。结果:最佳工艺条件为选用AB-8大孔吸附树脂,上样浓度36.32 mg·ml~(-1),最大上样量4倍柱床体积,以蒸馏水8倍柱床体积洗脱,洗脱流速为1.0 ml·min~(-1)。通过本工艺纯化的黄精小分子糖保留率达97.56%,纯度达90.93%。结论:大孔吸附树脂对黄精小分子糖有较好的纯化作用,该工艺简单可行,适合于工业化生产。
Objective: To study the optimum conditions and parameters for the purification of macroporous resin by macroporous resin. Methods: The retention rate and purity of small molecule sugar were investigated. The adsorption properties of small macromolecules of macroporous resin with four kinds of macroporous resins were compared. The effects of loading concentration, loading volume, elution flow rate and the amount of eluent were investigated. Resin purification process to determine the best process parameters. Purification by this process Polygonatum small molecular sugar retention rate of 97.56%, purity of 90.93%. Conclusion: The macroporous resin has a good purification effect on the polygalacturonase. The method is simple and feasible and suitable for industrial production.
