目的研究中药扶正汤对MRL/1Pr红斑狼疮小鼠肾损害相关细胞因子γ干扰素(IFN-γ)和细胞间粘附分子-1(ICAM-1)水平的影响。方法将MRL/1Pr模型小鼠随机分成4组:0.9%盐水对照组(A组)、西药组(B组)、中药组(C组)、中西医结合组(D组),治疗6周。取小鼠血清和肾组织匀浆应用酶联免疫法(ELISA)检测IFN-γ和ICAM-1水平。结果 3组治疗组血清中IFN-γ含量明显低于对照组(P<0.01),各治疗组间差异无统计学意义。肾组织中IFN-γ含量中药组较对照组、西药组和中西医结合组明显减少(P<0.01),而对照组、西药组、和中西医结合组间差异无统计学意义。对照组血清和肾组织中ICAM-1水平较高,3组治疗组ICAM-1水平均明显低于对照组(P<0.01),中药组ICAM-1水平较低,明显低于西药组和中西药组(P<0.01)。结论扶正汤可能通过下调细胞因子IFN-γ和ICAM-1水平,减少对肾组织的损伤,对狼疮小鼠肾炎发挥明显的治疗作用。
Objective To study the effects of Fuzheng Decoction on the expression of IFN-γ and ICAM-1 in renal injury of MRL / 1Pr lupus erythematosus mice. Methods The MRL / 1Pr model mice were randomly divided into 4 groups: 0.9% saline control group (A group), Western medicine group (B group), Chinese medicine group (C group) and Integrative Medicine group (D group) for 6 weeks. The levels of IFN-γ and ICAM-1 in serum and renal homogenate of mice were determined by enzyme-linked immunosorbent assay (ELISA). Results The levels of IFN-γ in serum of the three treatment groups were significantly lower than those of the control group (P <0.01). There was no significant difference among the three treatment groups. Compared with control group, western medicine group and traditional Chinese and western medicine group, IFN-γ content in renal tissue was significantly decreased (P <0.01), but there was no significant difference between control group, western medicine group and traditional Chinese and Western medicine group. ICAM-1 levels in the serum and kidney of the control group were higher than those in the control group (P <0.01), while ICAM-1 levels in the three treatment groups were significantly lower than those in the control group (P <0.01) Western medicine group (P <0.01). Conclusion Fuzheng Decoction may play a significant therapeutic effect on glomerulonephritis by down-regulating the levels of cytokines IFN-γ and ICAM-1, reducing the damage to the kidney.
