为了研究极低密度脂蛋白受体结构与功能的关系 ,本文利用反转录聚合酶链反应技术从中国人心肌组织中克隆出极低密度脂蛋白受体的全长cDNA后 ,将其插入真核表达载体pcDNA3获得重组体pCD -VR。测序结果发现 ,这一源于中国人的极低密度脂蛋白受体基因cDNA的碱基序列与文献报道基本相符。反转录聚合酶链反应检测证实 ,转染pCD -VR的中国仓鼠卵巢细胞可有效表达极低密度脂蛋白受体mRNA。脂蛋白结合实验结果发现 ,转染的中国仓鼠卵巢细胞结合极低密度脂蛋白的能力明显增高。
In order to study the relationship between the structure and function of very low density lipoprotein receptor, we cloned the full-length cDNA of very low density lipoprotein receptor from Chinese myocardium using reverse transcription polymerase chain reaction Recombinant pCD-VR was obtained by using the nuclear expression vector pcDNA3. The sequencing results showed that the base sequence of this very low density lipoprotein receptor gene cDNA derived from Chinese basically corresponded with that reported in the literature. Reverse transcriptase-polymerase chain reaction confirmed that Chinese hamster ovary cells transfected with pCD-VR could effectively express very low density lipoprotein receptor mRNA. Lipoprotein binding assay results showed that transfected Chinese hamster ovary cells significantly increased their ability to bind very low density lipoprotein.
