目的观察氟中毒大鼠生长板Ihh、PTHrP mRNA及蛋白表达的影响,探讨Ihh信号通路在氟致软骨损伤中的可能作用。方法 32只Wistar大鼠按体重随机分成4组,对照组饮用自来水,染氟组分别饮用含氟50、100和150 mg/L的自来水,3个月后测定大鼠血清及骨组织氟含量,Real-time PT-PCR法检测Ihh、PTHrP mRNA表达情况,免疫组化法检测软骨组织中PTHrP蛋白表达情况。结果染氟组大鼠生长板中Ihh、PTHrP mRNA表达显著高于对照组,差异有统计学意义(P<0.05)。对照组大鼠生长板软骨细胞不表达或极弱表达PThrP,而氟中毒大鼠生长板静止层和肥大层软骨细胞细胞质中PTHrP蛋白为阳性表达,且随染氟剂量增加而升高。结论过量氟可能通过影响Ihh、PTHrP mRNA及蛋白表达水平,造成生长板软骨增殖、分化失衡。
Objective To investigate the effects of Ihh signaling pathway on the expression of Ihh and PTHrP mRNA and protein in the growth plate of fluorosis rats. Methods 32 Wistar rats were randomly divided into 4 groups according to body weight. Drinking water was used in the control group. Tap water containing 50, 100 and 150 mg / L fluoride was drank in the control group, and serum and bone tissue contents in the rats were measured after 3 months. Real-time PT-PCR method was used to detect the expression of Ihh and PTHrP mRNA. The expression of PTHrP in cartilage was detected by immunohistochemistry. Results The expression of Ihh and PTHrP mRNA in the growth plate of rats stained with Fluoride was significantly higher than that of the control group (P <0.05). In the control group, PThrP was not expressed or weakly expressed in growth plate chondrocytes, while PTHrP protein was positive in the resting layer of hypertrophic layer and hypertrophic chondrocytes in fluorosis rats, and increased with the increase of fluoride dose. Conclusion Excessive fluoride may affect the expression of Ihh, PTHrP mRNA and protein, resulting in proliferation and imbalance of growth plate cartilage.
