目的:探讨青蒿素对急性白血病K562细胞的增殖抑制作用及其作用机制。方法:以不同浓度的青蒿素作用于体外培养的K562细胞,MTT法检测细胞生长抑制率,应用流式细胞仪及Hoechst 33258荧光染色法观察细胞凋亡,TRAP-PCR-ELISA法检测细胞凋亡前后的端粒酶活性。结果:青蒿素可显著降低K562细胞端粒酶活性,抑制细胞生长,诱导细胞发生凋亡,并呈现出明显的量-效与时-效关系。结论:青蒿素能抑制K562细胞的生长并诱导细胞发生调亡,降低细胞端粒酶活性可能是其重要作用机制之一。
Objective: To investigate the inhibitory effect of artemisinin on the proliferation of acute leukemia K562 cells and its mechanism. Methods: K562 cells cultured in vitro were treated with different concentrations of artemisinin. The cell growth inhibition rate was determined by MTT assay. The apoptosis was observed by flow cytometry and Hoechst 33258 staining. The apoptosis of cells was detected by TRAP-PCR-ELISA Telomerase activity before and after death. Results: Artemisinin could significantly reduce telomerase activity, inhibit cell growth and induce apoptosis in K562 cells, and showed a dose - effect and time - effect relationship. CONCLUSION: Artemisinin can inhibit the growth of K562 cells and induce the apoptosis of cells and reduce the telomerase activity may be one of the important mechanisms.
