目的:研究肠复方对裸鼠肠癌原位移植瘤的抑制作用。方法:采用瘤块接种法建立裸鼠大肠癌原位移植瘤模型。将造模成功的100只裸鼠按随机数字表法分入模型组(A组)、肠复方低剂量组(B组)、肠复方中剂量组(C组)、肠复方高剂量组(D组)和氟尿嘧啶(5-Fu)组(E组),每组20只,各组内随机分为两组(A1、A2、B1、B2、C1、C2、D1、D2、E1、E2组)。模拟临床给药,分别干预4周。干预过程中记录裸鼠体重变化并观察裸鼠生长状态。A1、B1、C1、D1、E1组干预结束后处死裸鼠,采集标本,记录瘤质量,计算抑瘤率和肝转移率。A2、B2、C2、D2、E2组继续饲养,记录荷瘤裸鼠生存时间。结果:肠复方各剂量组裸鼠生长状态明显优于模型组和5-FU组;肠复方各剂量组和5-FU组裸鼠体质量优于模型组,有统计学差异(P<0.01,P<0.05);肠复方各剂量组和5-FU组瘤质重低于模型组,差异有统计学差异(P<0.01,P<0.05);肠复方各剂量组小鼠平均生存时间明显高于模型组,差异有统计学意义(P<0.01,P<0.05);肠复方高、中剂量组及5-FU组肝转移抑瘤率明显高于模型组,差异有统计学意义(P<0.05)。结论:中药肠复方可以稳定患瘤裸鼠体质量、改善生长状态、降低瘤质量,同时中药肠复方能够延长患瘤裸鼠生存时间,降低肝转移率。
Objective: To study the inhibitory effect of Changfu Decoction on orthotopic transplanted colon cancer in nude mice. Methods: Nude mice model of orthotopic implantation of colorectal cancer was established by tumor inoculation. A total of 100 nude mice were randomly divided into model group (A group), low dose intestine group (B group), middle dose group (C group), high dose intestine compound group (D Group A and group B were randomly divided into two groups (A1, A2, B1, B2, C1, C2, D1, D2, E1 and E2 groups) and 5 fluorouracil groups (group E) . Simulated clinical administration, respectively, for 4 weeks. The body weight of nude mice was recorded during the intervention and the growth of nude mice was observed. The mice in A1, B1, C1, D1 and E1 groups were sacrificed and the specimens were collected. The tumor mass was recorded and the tumor inhibition rate and liver metastasis rate were calculated. A2, B2, C2, D2, E2 continue feeding group, record the survival time of tumor-bearing nude mice. Results: The growth of nude mice in each dose group was obviously better than that in model group and 5-FU group. The body weight of nude mice in each dose group and 5-FU group was significantly higher than that in model group (P <0.01, P <0.05). The tumor weight of each dose group and the 5-FU group was significantly lower than that of the model group (P <0.01, P <0.05), and the average survival time of each dose group was significantly higher (P <0.01, P <0.05). The inhibitory rates of liver metastasis in high and medium dose group and 5-FU group were significantly higher than those in model group (P <0.01, P <0.05), and the difference was statistically significant 0.05). CONCLUSION: GSI can stabilize the body weight of tumor-bearing nude mice, improve the growth status and reduce the tumor mass. At the same time, Ganyu Fugan can prolong the survival time of the tumor-bearing nude mice and reduce the liver metastasis rate.
