目的通过检测凋亡抑制因子(Livin mRNA)和凋亡促进因子(SMAC蛋白)在人肝细胞癌组织中的表达及相关性,分析其与肝细胞癌转移、复发的关系。方法通过反转录聚合酶链反应(RT-PCR)技术检测50例肝细胞癌和癌旁组织中Livin mRNA的表达;用免疫组化方法检测50例肝细胞癌和癌旁组织中SMAC蛋白表达水平。结果50例肝细胞癌组织中SMAC阳性率明显低于癌旁组织(P<0.05)。Livin mRNA和SMAC的表达与门脉癌栓、术后复发、肝外转移及肿瘤细胞分化密切相关(P<0.05);SMAC的表达还与临床分期密切相关(P<0.05)。Livin mRNA表达与SMAC蛋白表达呈显著负相关(P<0.05)。结论Livin mRNA的高表达及SMAC的失活可能在肝细胞癌的发生及进展中起着重要作用。
Objective To detect the expression and correlation of Livin mRNA and SMAC protein in human hepatocellular carcinoma (HCC) and to analyze the relationship between Livin mRNA and metastasis and recurrence of HCC. Methods The expression of Livin mRNA was detected by reverse transcriptase-polymerase chain reaction (RT-PCR) in 50 cases of hepatocellular carcinoma and adjacent non-cancerous tissues. The expressions of SMAC protein in 50 cases of hepatocellular carcinoma and adjacent normal tissue were detected by immunohistochemistry Level. Results The positive rate of SMAC in 50 cases of hepatocellular carcinoma was significantly lower than that in adjacent tissues (P <0.05). The expression of Livin mRNA and SMAC was closely related to portal vein tumor thrombus, postoperative recurrence, extrahepatic metastasis and tumor cell differentiation (P <0.05). The expression of SMAC was also closely related to clinical stage (P <0.05). Livin mRNA expression and SMAC protein expression was significantly negatively correlated (P <0.05). Conclusion The high expression of Livin mRNA and the inactivation of SMAC may play an important role in the occurrence and progression of hepatocellular carcinoma.
