目的探讨抗CD94单抗与NK细胞、γδ+T细胞及CTL的相关性。方法采用荧光检测法,分别分析NK细胞、γδ+T细胞和CTL上CD94分子的表达。采用3HTdR掺入法和MTT释放法,分析抗94单抗对这3种细胞的杀伤活性和细胞增殖的影响。结果NK细胞和γδ+T细胞上CD94分子的表达,明显高于PBL和CTL。抗CD94单抗能激活NK细胞、γδT细胞和CTL的增殖,并能提高γδ+T细胞和CTL的杀伤活性。结论CD94分子在NK细胞和γδ+T细胞表面呈高表达。抗CD94单抗对NK细胞、γδ+T细胞及CTL的增殖具有上调作用;对它们的杀伤活性具有正向调节作用。
Objective To investigate the relationship between anti-CD94 monoclonal antibody and NK cells, γδ + T cells and CTL. Methods The expression of CD94 on NK cells, γδ + T cells and CTL were analyzed by fluorescence detection. 3H-TdR incorporation and MTT release assay were used to analyze the effect of anti-94 mAb on the killing activity and cell proliferation of these three kinds of cells. Results The expression of CD94 on NK cells and γδ + T cells was significantly higher than that of PBL and CTL. Anti-CD94 mAb can activate the proliferation of NK cells, γδT cells and CTL, and enhance the killing activity of γδ + T cells and CTL. Conclusion CD94 molecules are highly expressed on the surface of NK cells and γδ + T cells. Anti-CD94 monoclonal antibody has an upregulation effect on the proliferation of NK cells, γδ + T cells and CTLs, and has a positive regulatory effect on their cytotoxic activity.
