目的研究弗氏枸橼酸杆菌CF74的T6SS基因组岛功能。方法构建弗氏枸橼酸杆菌CF74的T6SS基因组岛的精确缺失突变株(CF74ΔT6SS)。对弗氏枸橼酸杆菌CF74和其突变株CF74ΔT6SS的全菌蛋白和上清蛋白进行提取,并进行2-DE分析。结果在CF74ΔT6SS全菌蛋白中,有4个蛋白表达上调,42个蛋白表达下调,并且Fli C的表达下调十分明显;在CF74ΔT6SS的培养上清中,有27个蛋白表达上调,36个蛋白表达下调,并且Fli C、Flg K和Fli D的表达下调十分明显。结论弗氏枸橼酸杆菌CF74的T6SS可以影响Fli C蛋白的表达和分泌。
Objective To study the T6SS genomic island function of Citrobacter freundii CF74. Methods An exact deletion mutant (CF74ΔT6SS) of the T6SS genomic island from C. citrifaciens CF74 was constructed. The whole bacteria protein and the supernatant protein of Citrobacter freundii CF74 and its mutant CF74ΔT6SS were extracted and subjected to 2-DE analysis. Results In CF74ΔT6SS whole-cell protein, 4 proteins were upregulated, 42 proteins were down-regulated, and the expression of Fli C was down-regulated. In the CF74ΔT6SS culture supernatant, 27 proteins were up-regulated and 36 proteins were down-regulated , And the down-regulation of Fli C, Flg K and Fli D was evident. Conclusion T6SS of C. freundii CF74 can affect the expression and secretion of Fli protein.
