目的研究在大鼠肾移植中骨髓干细胞移植对免疫耐受的诱导作用及机制。方法40只Wistar大鼠(雌雄各半)随机分为实验组和对照组,进行肾移植手术,术前经门静脉分别注射骨髓细胞及生理盐水,30天后观察肉眼和光镜下排斥反应及检测嵌合体的形成。结果实验组肉眼和光镜下排斥反应均较轻微,嵌合体检测为阳性。结论应用他克莫司并注射供体骨髓细胞,可诱导一定的免疫耐受。移植前输注供体骨髓细胞而形成的嵌合体与免疫耐受有关。
Objective To investigate the effects of bone marrow stem cell transplantation on immune tolerance in rat kidney transplantation and its mechanism. Methods Forty Wistar rats (male and female, half) were randomly divided into experimental group and control group for renal transplantation. Bone marrow cells and normal saline were injected through the portal vein before operation. After 30 days, macroscopic and photomicroscopic rejection and chimerism Formation. Results In the experimental group, the rejection in both eyes and light microscopy was slight, and the chimera was positive. Conclusion Tacrolimus and injection of donor bone marrow cells can induce certain immune tolerance. Transplantation of donor bone marrow cells before transplantation and the formation of chimeras and immune tolerance.
