目的研究青蒿素联合琥珀酸亚铁对人肝癌细胞SMMC-7721的选择性抑制作用。方法将人肝癌细胞株SMMC-7721接种于小鼠腰部皮下,制成小鼠荷瘤动物模型,分别用青蒿素、琥珀酸亚铁、青蒿素+琥珀酸亚铁、生理盐水(对照组)对荷瘤小鼠进行干预,观察小鼠肿瘤体积的变化;TUNEL法检查肿瘤细胞的凋亡,计算抑瘤率和凋亡指数。结果青蒿素和青蒿素+琥珀酸亚铁与生理盐水相比,明显抑制荷瘤小鼠的肿瘤生长(P<0.05),其抑瘤率分别为33.21%和55.04%,2组间存在明显差异(P<0.05)。青蒿素联合琥珀酸亚铁表现出明显的促凋亡作用,凋亡指数为31.35%,与对照组相比差异有统计学意义(P<0.05)。结论青蒿素和青蒿素联合琥珀酸亚铁对人肝癌细胞SMMC-7721均有肿瘤抑制作用和促进肿瘤细胞凋亡的作用,但是联合用药作用更强。
Objective To study the selective inhibitory effect of artemisinin combined with ferrous succinate on SMMC-7721 cells. Methods The human hepatocellular carcinoma cell line SMMC-7721 was inoculated subcutaneously in the lumbar region of the mouse, and a mouse tumor-bearing animal model was made. Artemisinin, ferrous succinate, artemisinin + ferrous succinate, and normal saline (control group) were used. ) Tumor-bearing mice were intervened to observe the changes in the tumor volume of the mice; TUNEL assay was used to examine the apoptosis of tumor cells, and tumor inhibition rate and apoptosis index were calculated. Results Artemisinin, artemisinin and ferrous succinate significantly inhibited the tumor growth of tumor-bearing mice compared with normal saline (P<0.05). The tumor inhibition rates were 33.21% and 55.04%, respectively, between the two groups. Significant difference (P<0.05). Artemisinin combined with ferrous succinate showed a significant pro-apoptotic effect with an apoptotic index of 31.35%, which was significantly different from that of the control group (P<0.05). Conclusion Artemisinin and artemisinin combined with ferrous succinate can inhibit tumor cells and promote apoptosis of human hepatoma SMMC-7721 cells, but the effect of combination therapy is stronger.
