本文采用反相色谱法及固相酶柱后反应技术,建立了高灵敏和特异性的人体液结合型胆汁酸测定方法;研究了最佳酶反应和酶活力保存条件,方法回收率88~99%,重复性cv1.2~3.7%(n=9),检测线性范围0~500ng,最低检知量12~23ng,固相酶柱柱寿命长达6个月。用本法测定35例人胃液标本,比较了患者胃液胆汁酸与正常人的差异。
In this paper, high-sensitivity and specific bile acid determination methods were established using reversed-phase chromatography and solid phase post-column post-reaction technology. The optimal enzyme reaction and enzyme activity preservation conditions were studied. The recovery rate of the method was 88~99. %, reproducibility of cv1.2 ~ 3.7% (n = 9), detection linear range of 0 ~ 500ng, the minimum detection amount of 12 ~ 23ng, solid phase enzyme column life of up to 6 months. Thirty-five human gastric fluid specimens were measured by this method and the differences between patients’ bile acids in gastric juice and normal persons were compared.
