目的探讨乳腺癌组织中α-1,6岩藻糖基转移酶mRNA和蛋白的表达水平及其临床意义。方法应用蛋白免疫印迹法、实时荧光定量PCR法检测40例乳腺癌患者术后的癌组织、癌旁组织以及40名健康者的正常乳腺组织中α-1,6岩藻糖基转移酶在的表达情况,并分析其与乳腺癌之间的关系。结果 FUT8 mRNA和蛋白在乳腺癌组织中的表达水平高于癌旁组织和正常乳腺组织,差异有统计学意义(P<0.05);Ⅲ期乳腺癌组织中FUT8蛋白表达水平明显高于Ⅰ-Ⅱ期,差异有统计学意义(P<0.001);有淋巴结转移的乳腺癌组织中FUT8蛋白表达水平明显高于无淋巴结转移组,差异有统计学意义(P<0.001)。结论高FUT8蛋白的表达与乳腺癌密切相关,其可能的调控机制及生物学效应有待进一步研究。
Objective To investigate the expression of α-1,6 fucosyltransferase mRNA and protein in breast cancer and its clinical significance. Methods Western blotting and real-time fluorescence quantitative PCR were used to detect the expression of α-1,6 fucosyltransferase in 40 cases of breast cancer tissues, adjacent tissues and 40 normal breast tissues Express the situation and analyze its relationship with breast cancer. Results The expression of FUT8 mRNA and protein in breast cancer tissues was significantly higher than that in paracancerous tissues and normal breast tissues (P <0.05). The expression of FUT8 protein in stage Ⅲ breast cancer tissues was significantly higher than that in stage Ⅰ-Ⅱ (P <0.001). The expression of FUT8 protein in breast cancer with lymph node metastasis was significantly higher than that without lymph node metastasis, the difference was statistically significant (P <0.001). Conclusion The expression of high FUT8 protein is closely related to breast cancer. The possible regulatory mechanisms and biological effects need to be further studied.
