目的探讨脂联素基因SNP+45T/G单核苷酸多态性与妊娠期糖尿病(GDM)及其妊娠结局的相关性。方法采用PCR-限制性内切酶片段长度多态性(PCR-RFLP)技术检测158例GDM孕妇(GDM组)、128例妊娠期糖耐量异常(GIGT组)及120例正常孕妇(对照组)的脂联素基因SNP+45T/G多态性,分析脂联素基因多态性与相应新生儿结局的关系。结果南通地区汉族人群中基脂联素因+45位点有T/G单核苷酸多态性存在。三组的基因型无统计学差异(P>0.05),但GDM组和GIGT组的G等位基因频率高于对照组(P<0.05)。携带G等位基因孕妇的新生儿低血糖、巨大儿及新生儿窒息的发生率明显高于阴性表达组(P<0.05)。结论脂联素基因SNP+45T/G多态性与GDM的发生存在一定相关性,且与其妊娠结局有关。
Objective To investigate the association between the SNP + 45T / G single nucleotide polymorphism of adiponectin gene and gestational diabetes mellitus (GDM) and its pregnancy outcome. Methods 158 GDM pregnant women (GDM group), 128 gestational impaired glucose tolerance (GIGT group) and 120 normal pregnant women (control group) were detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) Of adiponectin gene SNP + 45T / G polymorphism, analysis of adiponectin gene polymorphisms and the corresponding neonatal outcome. Results There was T / G SNP at the +45 locus of adiponectin in Nantong Han population. The genotypes of the three groups had no significant difference (P> 0.05), but the frequencies of G allele in GDM group and GIGT group were higher than those in control group (P <0.05). Neonatal hypoglycemia, maternal and neonatal asphyxia in pregnant women with G allele were significantly higher than those in the negative expression group (P <0.05). Conclusion The SNP + 45T / G polymorphism of adiponectin gene is associated with the occurrence of GDM and related to its pregnancy outcome.
